119 Noncanonical NF-κB signaling is essential for development of chronic contact hypersensitivity (CHS)

Noncanonical NF-κB signaling(NF-κB2) plays an essential role in adaptive immunity. Aim of our study was to uncover the impact NF-κB2 signaling on T cell mediated delayed-type hypersensitivity reactions skin by investigating acute and chronic TNCB-induced CHS, which represents a prototypic model autoimmune disease. We conducted extensive vivo PET/MRI studies ex analyses NF-κB2-/- wild type(WT) mice impairments immune migration as well differences inflammatory microenvironment. Impairment did not affect phase no inflammation infiltration between WT were observed. However, inflamed ears with CHS displayed significantly reduces ear swelling responses strongly reduced cells into draining lymph nodes when compared mice. Histopathological analysis tissue revealed lack edema, acanthosis hyperkeratosis, along cells, neutrophils macrophages. Further, proteome from showed heavily expression chemokines CXCL2, CXCL5, CXCL9, CXCL10, CCL11 CCL12 angiogenesis promoting mediators MMP9, HGF, FGF AREG. Moreover, exhibited endothelial adhesion molecules ICAM-1 E-selectin. In conclusion, disruption alleviates impairing chemotaxis, resulting decreased recruitment site inflammation.
